Biological Background: CD45R/B220 Function and Localization

  • Receptor-type tyrosine-protein phosphatase C (PTPRC), commonly known as CD45, L-CA, Ly-5, or T200, is a transmembrane glycoprotein. The B220 isoform is specifically expressed on B cells and corresponds to the high-molecular-weight variant of CD45.
  • CD45 is essential for T-cell activation through the T-cell receptor (TCR), acting as a positive regulator of T-cell coactivation upon binding to DPP4. The first phosphatase domain carries enzymatic activity, while the second domain influences substrate specificity.
  • Upon T-cell activation, CD45 recruits and dephosphorylates SKAP1 and FYN, and also dephosphorylates LYN, thereby modulating their kinase activities.
  • At the immunological synapse, CD45 interacts with CLEC10A on immature dendritic cells; this interaction recognizes Tn antigen-bearing CD45 on effector T cells and contributes to the modulation of T-cell activation thresholds, helping to prevent autoreactivity.
  • CD45 localizes to the cell membrane, membrane rafts, and the immunological synapse, consistent with its role in signal transduction during cell–cell contact.
  • Alternative splicing generates multiple CD45 isoforms (e.g., CD45R, CD45RO, B220), which are differentially expressed among leukocyte subpopulations and influence signaling outcomes.
  • Post-translational modifications include extensive N- and O-linked glycosylation and phosphorylation, which regulate phosphatase activity and surface expression.
  • The mouse CD45 is encoded by the Ptprc (Ly-5) gene. The antibody described here cross-reacts with mouse CD45R/B220.

Experimental Guidance and Technical Tips

  • The antibody is conjugated to , a violet-laser-excitable fluorophore. Verify that your flow cytometer is equipped with a 405 nm laser and appropriate emission filters (e.g., 450/50 bandpass) for optimal detection.
  • According to the provider, the antibody cross-reacts with mouse CD45R/B220. If working with human or other species’ samples, perform appropriate cross-reactivity tests before use.
  • CD45R/B220 is predominantly expressed on B cells; include well-characterized positive (e.g., splenocytes or B-cell lines) and negative (e.g., T cells) staining controls to confirm specificity in flow cytometry panels.
  • The immunogen used for generation includes a peptide from mouse/human CD45R/B220; thus, the antibody could theoretically recognize human CD45R/B220. Validation in the relevant species and sample system is recommended.
  • Protein weights of 130–144 kDa represent different CD45 isoforms. For applications beyond flow cytometry (e.g., Western blotting), ensure appropriate molecular weight markers and consider that this product is validated only for FC.
  • Store and handle the antibody as recommended for fluorescent conjugates; upon receipt, the product is shipped on ice bags to maintain stability.
