  • CD45R/B220, encoded by the Ptprc gene, is the mouse homolog of human CD45, also known as leukocyte common antigen (L-CA) or Ly-5. It exists in multiple isoforms generated by alternative splicing, with molecular weights of approximately 130, 140, and 144 kDa.
  • Functions as a receptor-type tyrosine-protein phosphatase, essential for T-cell activation through the antigen receptor. Acts as a positive regulator of T-cell coactivation upon binding to DPP4.
  • The protein contains two phosphatase domains: the first has enzymatic activity, while the second modulates substrate specificity. Upon T-cell activation, it recruits and dephosphorylates signaling molecules such as LYN, SKAP1, and FYN.
  • CD45R/B220 is localized to the cell membrane, membrane rafts, and immunological synapse, consistent with its role in modulating immune cell signaling at sites of cell-cell contact.
