Biological Background: CD45R/B220 (Receptor-type tyrosine-protein phosphatase C) Function and Localization
- CD45R/B220, encoded by the Ptprc gene, is a receptor-type tyrosine-protein phosphatase C (PTPRC), also known as leukocyte common antigen (L-CA), lymphocyte antigen 5 (Ly-5), and T200. Multiple isoforms are generated through alternative splicing, producing variants of different molecular weights (e.g., 130, 140, 144 kDa).
- PTPRC functions as a critical positive regulator of T-cell activation: its first phosphatase domain has enzymatic activity, while the second one modulates substrate specificity. Upon T-cell receptor engagement, it recruits and dephosphorylates signaling molecules such as SKAP1 and FYN (by similarity).
- It also dephosphorylates the Src-family kinase LYN, thereby modulating LYN kinase activity and influencing downstream signaling cascades in lymphocytes.
- Subcellularly, PTPRC is localized to the cell membrane, enriched in membrane lipid rafts, and concentrates at the immunological synapse between T cells and antigen-presenting cells.
- PTPRC interacts with CLEC10A on immature dendritic cells at the T cell–dendritic cell interface; this interaction recognizes Tn antigen-carrying CD45 on effector T cells, adjusting the threshold for T cell activation to limit autoreactivity.
