Biological Background: CD3 (delta and epsilon chains) Function and Localization
- CD3 delta (gene Cd3d) and epsilon (Cd3e) are essential components of the T-cell receptor (TCR)-CD3 complex on the surface of T lymphocytes, playing a central role in adaptive immunity.
- The CD3 complex (delta, epsilon, gamma, and CD247/zeta chains) transduces activation signals upon TCR engagement by antigen-presenting cells, via phosphorylation of immunoreceptor tyrosine-based activation motifs (ITAMs) in their cytoplasmic domains.
- CD3D is critical for thymocyte differentiation and proper surface expression of the TCR-CD3 complex; disruption impairs T-cell development. Related references: PMID:10935641
- CD3 delta interacts with coreceptors CD4 and CD8, functionally linking TCR recognition to co-receptor signaling, which is required for activation and positive selection of CD4+ or CD8+ T cells.
- Both CD3 delta and epsilon chains are type I transmembrane glycoproteins localized to the plasma membrane.
- Key post-translational modifications include disulfide bond formation, glycosylation, and ITAM phosphorylation by Src family kinases LCK and FYN.
- Together, CD3 delta and epsilon chains have molecular weights of approximately 19 kDa and 21 kDa, respectively.
Experimental Guidance and Technical Tips
- The fluorescent conjugate enables direct detection in flow cytometry (FC) without the need for secondary staining reagents.
- To reduce non-specific binding, consider pre-incubating cells with an appropriate Fc receptor blocking reagent and use a matched rat IgG isotype control.
- Optimize antibody concentration by titration over a range (e.g., 0.1–5 µg/mL) in the relevant cell system to achieve the best signal-to-noise ratio.
- Prepare single-cell suspensions and assess viability before staining to ensure robust and reproducible results.
- The antibody is recommended for use with mouse samples; validate assay conditions for your specific experimental setup.
