Biological Background: Raptor Function and Localization
- Raptor (Regulatory-associated protein of mTOR, also known as p150 TOR-scaffold protein) is a key component of the mTORC1 complex, serving as both a molecular adapter for lysosomal recruitment and a substrate-specific adapter that directs TOS motif-containing proteins to the MTOR kinase domain for phosphorylation. - mTORC1 is an evolutionarily conserved central nutrient sensor that stimulates anabolic reactions and macromolecule biosynthesis, promoting protein, lipid, and nucleotide synthesis via phosphorylation of substrates like RPS6KB1 and EIF4EBP1. - For Western blot, a band around 149 kDa is expected; consider using appropriate gel percentage and transfer conditions for high-molecular-weight proteins.
- For ELISA, the antibody can be used as capture or detection reagent; pairing with another clone may be needed for sandwich assays.
