Biological Background: RanGAP1 Function and Localization
- RanGAP1 (Ran GTPase-activating protein 1) serves as a GTPase activator for the small GTPase RAN, accelerating the conversion of GTP-bound RAN to GDP-bound RAN in the cytoplasm, a critical step for Ran-dependent nuclear transport. - It mediates the dissociation of export cargo from nuclear export complexes that contain XPO1, RAN, and RANBP2 following nuclear export. Related references: PMID:27160050
- The protein localizes to multiple cellular compartments, including the cytoplasm, nucleoplasm, nuclear envelope, centromeres, kinetochores, and the mitotic spindle, reflecting its role in both interphase transport and mitosis.

- For Western blot analysis, a predominant band near 64 kDa is expected in human cell lysates or tissue extracts; consider including positive controls such as HeLa, HEK293, or testis lysates.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), validate the antibody in human tissues or cell lines with known RanGAP1 expression (e.g., testis, brain); optimization of antigen retrieval and permeabilization conditions is recommended.
- For ELISA applications, use recombinant RanGAP1 protein or a peptide derived from the immunogen sequence as a coating antigen to assess antibody specificity and titer.
