- RAD54L (also known as hHR54, hRAD54, RAD54A) is a multifunctional ATPase that plays a central role in homologous recombination (HR), a major pathway for repairing DNA double-strand breaks (DSBs), single-stranded DNA gaps, and stalled or collapsed replication forks.

- Essential for the formation of RAD51-mediated synaptic complexes, a three-stranded nucleoprotein filament that stabilizes homologous pairing once sequence identity is recognized.
- After strand exchange, RAD54L promotes the dissociation of RAD51 from dsDNA, facilitating the completion of the recombination cycle.
- Localized predominantly in the nucleus, consistent with its DNA repair function.
