Biological Background: SPDYA Function and Localization
- Regulates the G1/S phase transition of the cell cycle by binding and activating CDK1 and CDK2. Related references: PMID:12972555
- Contributes to CDK2 activation without promoting CDK2 phosphorylation by inducing a conformational change in the CDK2 T-loop that obstructs the substrate-binding cleft prior to kinase activation. Related references: PMID:28666995
- Mediates cell survival during the DNA damage process through activation of CDK2. Related references: PMID:12839962
- Subcellularly localized to the nucleus.
- Highly expressed in testis, with lower expression in a wide range of tissues including bone marrow, brain, heart, kidney, colon, liver, placenta, spleen, skeletal muscle, and others; expression is cell cycle-dependent and restricted to the G1/S phase.
- Subject to alternative splicing and phosphorylation (phosphoprotein), suggesting the existence of multiple isoforms and post-translational regulation.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein covering the full-length human SPDYA (amino acids 1-313), enabling recognition of both native and denatured protein forms.
- For Western blot, expect a band around 36 kDa; optimize gel percentage and transfer conditions accordingly.
- Cross-reactivity with mouse and rat SPDYA is noted; verify performance in your specific sample system.
- For ELISA, consider optimizing coating buffer and antibody dilution; both direct and indirect ELISA protocols can be applied.
