Biological Background: CD4 Function and Localization
- CD4, also known as T-cell surface antigen T4/Leu-3 and W3/25 antigen in rat, is a cell surface glycoprotein and a member of the immunoglobulin superfamily.
- Functions as a coreceptor for MHC class II molecules on antigen-presenting cells, facilitating T-cell receptor (TCR) signaling by recruiting the tyrosine kinase LCK to the TCR-CD3 complex.
- Essential for thymic development of T-helper cells and triggers differentiation of monocytes into mature macrophages.
- In macrophages and natural killer (NK) cells, CD4 contributes to differentiation, activation, cytokine expression, and cell migration via TCR/LCK-independent pathways.
- Encoded by the Cd4 gene in rat (Gene ID: 24933; human ortholog Gene ID: 920).
- Localized to the cell membrane as a single-pass type I membrane protein.
- Post-translational modifications include disulfide bond formation, N-glycosylation, and palmitoylation, which regulate its membrane localization and function.
