Biological Background: CD31/PECAM1 Function and Localization

  • Platelet endothelial cell adhesion molecule (PECAM-1/CD31) is a transmembrane glycoprotein belonging to the immunoglobulin superfamily, encoded by the Pecam1 gene.
  • Functions as a cell adhesion molecule essential for leukocyte transendothelial migration (TEM) under inflammatory conditions; Tyr-679 is critical for TEM by regulating PECAM1 trafficking to the lateral border recycling compartment (LBRC).
  • Mediates homophilic endothelial cell-cell adhesion at junctions and heterophilic interaction with CD177 to facilitate neutrophil transmigration.
  • Prevents macrophage-mediated phagocytosis of viable leukocytes via homophilic ligation, while promoting phagocytosis of apoptotic leukocytes by tethering them to phagocytes and disabling the detachment signal.
  • Modulates bradykinin receptor BDKRB2 activation and regulates bradykinin- and hyperosmotic shock-induced ERK1/2 signaling in endothelial cells.
  • Induces susceptibility to atherosclerosis (PubMed:19048083). Related references: PMID:19048083
  • Subcellularly located at the cell membrane, membrane rafts, and cell junctions.
  • Undergoes alternative splicing, yielding isoforms with predicted molecular weights of 69–81 kDa; also subject to palmitoylation and phosphorylation.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the extracellular domain (aa 18–220), suggesting the antibody recognizes an epitope in the N-terminal region, likely accessible for flow cytometry on live cells.
  • For flow cytometry, consider titrating the antibody to determine optimal staining concentration, and include appropriate isotype controls and Fc receptor blocking.
  • Validate in relevant sample systems such as mouse endothelial cells or platelets, where CD31 is highly expressed.
  • Note that the antibody reacts with both mouse and rat CD31, enabling cross-species applications; verify cross-reactivity with appropriate controls.
