Biological Background: CD31/PECAM1 Function and Localization
- CD31 (PECAM-1) is a platelet endothelial cell adhesion molecule belonging to the immunoglobulin superfamily, involved in cell-cell adhesion.
- It plays a critical role in leukocyte transendothelial migration (TEM); phosphorylation at Tyr-679 is essential for trafficking to the lateral border recycling compartment (LBRC).
- Homophilic interactions mediate endothelial cell junction integrity, while heterophilic binding to CD177 facilitates neutrophil transendothelial migration.
- PECAM-1 regulates macrophage phagocytosis: it promotes engulfment of apoptotic leukocytes while preventing phagocytosis of viable cells through a detachment signal.
- It modulates bradykinin receptor BDKRB2 activation and regulates bradykinin- and hyperosmotic shock-induced ERK1/2 activation in endothelial cells.
