Biological Background: CD27 Function and Localization
- CD27 (TNFRSF7) is a member of the tumor necrosis factor receptor superfamily, serving as a costimulatory immune-checkpoint receptor.
- Engagement by its ligand CD70 on B-cells triggers T-cell activation and expansion, promoting immune surveillance (PubMed:11062504). Related references: PMID:11062504
- CD70 ligation activates the TRAF2-PTPN6 signaling axis, which inhibits LCK phosphorylation to facilitate T-cell memory formation (By similarity).
- In early myeloid progenitor cells, CD70-CD27 signaling provides negative feedback to modulate hematopoiesis (PubMed:15723067). Related references: PMID:15723067
- CD27 negatively regulates the function of Th2 lymphocytes in adipose tissue (PubMed:38386557). Related references: PMID:38386557
- Subcellular localization: Cell membrane; single-pass type I transmembrane protein.
- Tissue specificity: Expressed in thymus, spleen, and on myeloid progenitor cells; not detected in non-lymphoid tissues (PubMed:8384562). Related references: PMID:8384562
- The mature protein has a predicted molecular weight of ~28 kDa and contains characteristic extracellular cysteine-rich repeats, a transmembrane helix, and a cytoplasmic tail. It is glycosylated and contains disulfide bonds (UniProt: P41272).
Experimental Guidance and Technical Tips
- This antibody recognizes an extracellular epitope (aa 24-182) and is suitable for native cell surface staining in flow cytometry. Titrate the antibody on a positive control cell line or primary cells (e.g., mouse splenocytes) to determine the optimal working concentration.
- Include appropriate viability dyes or fixable live/dead markers to exclude dead cells from analysis.
- Validate staining in the relevant sample system, and consider Fc receptor blocking if non-specific binding is a concern.
