Biological Background: CD27 Function and Localization

  • CD27 is a costimulatory immune checkpoint receptor belonging to the tumor necrosis factor receptor superfamily (TNFRSF7), also known as CD27L receptor or T-cell activation antigen CD27.
  • It is primarily expressed on the surface of T-cells, NK-cells, and B-cells, where it binds to its ligand CD70/CD27L to mediate antigen-specific T-cell activation and expansion, providing immune surveillance of B-cells. Related references: PMID:11062504
  • CD70-CD27 signaling activates the TRAF2-PTPN6 axis, which inhibits LCK phosphorylation, promoting T-cell memory formation.
  • Beyond T costimulation, CD27 on early myeloid progenitor cells provides a negative feedback signal regulating hematopoiesis during immune activation. Related references: PMID:15723067
  • CD27 also negatively regulates Th2 lymphocyte function in adipose tissue. Related references: PMID:38386557
  • Subcellularly, CD27 localizes to the cell membrane as a single-pass transmembrane receptor.
  • It undergoes post-translational modifications including glycosylation and disulfide bond formation, which are essential for its structure and function.
  • Tissue expression is restricted to lymphoid organs, notably thymus and spleen, and on myeloid progenitor cells. Related references: PMID:8384562

Experimental Guidance and Technical Tips

  • This antibody is provided as an conjugate, suitable for direct flow cytometry (FC) staining without the need for secondary antibodies. Consider titrating the antibody to determine the optimal concentration for your specific cell type and staining conditions.
  • Since the immunogen comprises amino acids 24-182 of mouse CD27 (corresponding to the extracellular domain), the antibody likely recognizes an extracellular epitope, making it suitable for live-cell surface staining.
  • For multicolor FC panels, ensure appropriate compensation controls and use isotype-matched negative controls to correct for non-specific binding.
  • Verify cross-reactivity in rat samples by testing on known positive and negative cells, as cross-reactivity is reported but may require individual validation.
  • CD27 is expressed on T-cells, NK-cells, and B-cells; thus, include activation markers or lineage markers to define cell subsets of interest.
