Biological Background: CD55/DAF Function and Localization
- Complement decay-accelerating factor (CD55/DAF) is a GPI-anchored membrane glycoprotein that regulates the complement cascade.
- It recognizes C4b and C3b fragments deposited on cell surfaces, interfering with the formation of C3 and C5 convertases (C4b2a and C3bBb) and thereby inhibiting complement amplification.
- CD55 prevents assembly and accelerates decay of the C3/C5 convertases, protecting host cells from autologous complement-mediated damage.
- The protein is anchored to the cell membrane via a glycosylphosphatidylinositol (GPI) moiety.
- It is expressed in brain, secretory epithelia, skeletal muscle, liver, testes, thymus, spleen, and lymph node.
- Contains Sushi (CCP/SCR) domains, disulfide bonds, and N-linked glycosylation sites.
