Biological Background: VISTA Function and Localization
- VISTA (V-type immunoglobulin domain-containing suppressor of T-cell activation), also known as Dies1, PD-1H, and Vsir, is a ~34 kDa transmembrane glycoprotein and member of the immunoglobulin superfamily (UniProt Q9D659).
- Functions as an immunoregulatory receptor that inhibits T-cell responses (PubMed:21383057, PubMed:24743150, PubMed:25267631). Related references: PMID:21383057 PMID:24743150 PMID:25267631
- May promote differentiation of embryonic stem cells by attenuating BMP4 signaling (PubMed:20042595). Related references: PMID:20042595
- By similarity, may stimulate MMP14-mediated MMP2 activation, highlighting a potential role in matrix remodeling.
- Predominantly localized to the cell membrane, with broad expression in hematopoietic tissues including spleen, thymus, bone marrow, and lymph nodes, as well as on CD4+ and CD8+ T cells, dendritic cells, macrophages, neutrophils, and NK cells (PubMed:21768399, PubMed:21383057). Related references: PMID:21768399 PMID:21383057
- Lower expression detected in non-hematopoietic tissues such as heart, brain, lung, kidney, muscle, ovary, and testis (PubMed:21383057, PubMed:21768399). Related references: PMID:21383057 PMID:21768399
- Contains an immunoglobulin V-set domain and is post-translationally modified by glycosylation and phosphorylation, with two disulfide bonds stabilizing its structure (Keywords).
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular region (aa 33-191) of mouse VISTA, consistent with the antibody’s use in live-cell flow cytometry. Consider titrating the antibody for optimal staining of mouse immune cells from spleen or lymph node.
- Validated for FC, but may also be suitable for other applications such as immunofluorescence or functional blocking studies; however, these require independent validation.
- Given the expression pattern, include appropriate isotype controls and Fc block when analyzing mouse leukocyte subsets.
