Biological Background: VISTA Function and Localization
- V-type immunoglobulin domain-containing suppressor of T-cell activation (VISTA), also known as Dies1, PD-1H, or V-set domain-containing immunoregulatory receptor, is encoded by the Vsir gene in mice.
- Acts as an immunoregulatory receptor that inhibits the T-cell response. Related references: PMID:21383057, PMID:24743150, PMID:25267631
- May promote differentiation of embryonic stem cells by inhibiting BMP4 signaling. Related references: PMID:20042595
- May stimulate MMP14-mediated MMP2 activation, based on similarity evidence.
- Localizes to the cell membrane as a single-pass type I membrane protein.
- Expressed in spleen, thymus, bone marrow, lymph node, and T-cells; also detected at lower levels in non-hematopoietic tissues such as heart, brain, lung, kidney, muscle, ovary, and testis.
- Contains one immunoglobulin-like domain, a signal peptide, and a transmembrane helix; classified under keywords: Cell membrane, Disulfide bond, Glycoprotein, Immunoglobulin domain, Membrane, Phosphoprotein, Receptor, Reference proteome, Signal, Transmembrane, Transmembrane helix.
Experimental Guidance and Technical Tips
- The immunogen encompasses the extracellular domain (amino acids 33-191), suggesting strong suitability for detecting cell surface VISTA via flow cytometry.
- For flow cytometry applications, consider titrating the antibody to determine the optimal concentration and using appropriate isotype controls.
- Validate staining in relevant mouse tissues or cell lines known to express VISTA, such as spleen, thymus, or bone marrow-derived cells.
