Biological Background: VISTA Function and Localization
- VISTA (V-type immunoglobulin domain-containing suppressor of T-cell activation), also known as PD-1H, Dies1, or Gi24, is a transmembrane protein encoded by the Vsir gene.
- It functions as an immunoregulatory receptor that inhibits T-cell responses, playing a critical role in immune checkpoint regulation. Related references: PMID:21383057, PMID:24743150, PMID:25267631
- VISTA may also promote embryonic stem cell differentiation by inhibiting BMP4 signaling. Related references: PMID:20042595
- Additionally, it may stimulate MMP14-mediated MMP2 activation (By similarity).
- VISTA is a cell membrane protein, consistent with its role as a receptor, and contains an immunoglobulin domain, a signal peptide, and a transmembrane helix, with post-translational modifications including glycosylation and phosphorylation.
- It is expressed in a wide range of immune cells and tissues, including CD4+ and CD8+ T-cells, dendritic cells, macrophages, neutrophils, and NK cells, as well as in non-hematopoietic organs.
