Biological Background: S100A9 Function and Localization
- S100A9 (also known as calgranulin-B, MRP14, p14) is a member of the S100 family of calcium-binding proteins, with a molecular weight of approximately 13 kDa.
- S100A9 is a calcium- and zinc-binding protein that plays a prominent role in the regulation of inflammatory processes and immune response. Related references: PMID:15331440 PMID:17767165 PMID:18403730
- Intracellularly, as part of the calprotectin complex with S100A8, it facilitates leukocyte arachidonic acid trafficking and metabolism, modulates the tubulin-dependent cytoskeleton during migration of phagocytes, and activates the neutrophilic NADPH-oxidase. Related references: PMID:15331440
- Extracellularly, S100A9 exerts pro-inflammatory, antimicrobial, oxidant-scavenging, and apoptosis-inducing activities. Related references: PMID:21382888
- It acts as an alarmin, binding to TLR4 and AGER, which activates MAP-kinase and NF-kappa-B signaling pathways, amplifying the pro-inflammatory cascade. Related references: PMID:17767165 PMID:18403730 PMID:19402754
- S100A9 is localized in the cytoplasm, associated with the cytoskeleton, and can be secreted; it also localizes to the cell membrane.
- Post-translational modifications include acetylation, methylation, and phosphorylation, which regulate its diverse functions.
Experimental Guidance and Technical Tips
- For intracellular flow cytometry, fix and permeabilize cells according to standard protocols (e.g., using a fixation/permeabilization kit) to allow antibody access to cytoplasmic S100A9.
- Titrate the -conjugated antibody to determine the optimal concentration for your specific cell type and experimental conditions.
- Include appropriate positive controls (e.g., mouse neutrophils or monocytes, which express high levels of S100A9) and negative controls (isotype control or unstimulated cells) in your assay.
- Consider that S100A9 can form complexes with S100A8; this interaction may influence epitope accessibility, so validate if the antibody detects the monomeric or complexed form in your assay.
- This antibody was raised against the full-length mouse S100a9 sequence (amino acids 1-113), so it may cross-react with other species if sequences are highly conserved, but the supplier only validates mouse reactivity. Test in other species if needed.
