Biological Background: LAMP1/CD107a Function and Localization
- LAMP1/CD107a, also known as LGP-120, P2B, or CD107a, is a heavily glycosylated lysosomal membrane glycoprotein with a molecular weight of ~43 kDa.
- Plays a key role in lysosome biogenesis, autophagy, and cholesterol homeostasis, and helps regulate lysosomal pH by inhibiting the proton channel TMEM175 (PubMed:15121881). Related references: PMID:15121881
- In NK cells, LAMP1 is critical for cytotoxicity: it facilitates cytotoxic granule movement, perforin trafficking, and protects NK cells from degranulation-induced self-damage (PubMed:23847195). Related references: PMID:23847195
- Presents carbohydrate ligands to selectins, facilitating cell adhesion (By similarity).
- Implicated in tumor cell metastasis (PubMed:2676155). Related references: PMID:2676155
- Subcellular localization includes lysosomal membrane, late endosomes, endosomes, cell membrane, and cytolytic granule membrane.
- Post-translational modifications include disulfide bonds and extensive glycosylation; the protein contains a signal peptide and a single transmembrane helix.
Experimental Guidance and Technical Tips
- For immunofluorescence (IF/ICC), permeabilization with detergents like Triton X-100 or saponin is recommended to detect intracellular lysosomal LAMP1.
- For flow cytometry, cell surface LAMP1 may be detected on activated NK cells or following degranulation; validate expression in your experimental system.
- Use an appropriate isotype control and consider conjugated secondary antibodies if signal amplification is needed.
- The conjugate is compatible with FITC filter sets; ensure minimal fluorophore quenching during staining and imaging.
