Biological Background: Ki67 Function and Localization

  • Encoded by the Mki67 gene and also known as Ki-67, Antigen KI-67 homolog, or proliferation marker protein Ki-67, this large nuclear protein is expressed in actively cycling cells.
  • Associates with the surface of mitotic chromosomes and acts as a chromosome repellent during early mitosis, required to maintain individual chromosome dispersion after nuclear envelope breakdown (PubMed:27362226). Related references: PMID:27362226
  • During late mitosis, it switches to a chromosome attractant, promoting chromosome clustering and helping exclude large cytoplasmic particles from the future nuclear space.
  • Forms extended brush-like structures on the chromosome surface that provide a steric and electrostatic barrier; its high net charge acts as a surfactant, preventing chromosome collapse into a single mass (PubMed:27362226). Related references: PMID:27362226
  • Regulated by phosphorylation: dephosphorylation during mitotic exit exposes a conserved basic patch, triggering RNA-dependent liquid-liquid phase separation on chromosome surfaces to drive clustering.
  • Binds DNA with a preference for supercoiled and AT-rich regions, and also binds premature ribosomal RNAs during anaphase.
  • Localizes to nucleoli in interphase, relocates to the chromosome surface during mitosis; mainly present in proliferating cells.
  • Post-translational modifications include acetylation, phosphorylation, and ubiquitination, consistent with complex cell cycle regulation.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 1816–2163 of mouse Ki67, covering a C-terminal region; it may recognize full-length protein and potential isoforms.
  • For intracellular flow cytometry, ensure appropriate fixation and permeabilization protocols, as the target is nuclear.
  • Validate the antibody in your specific cell type and experimental context; Ki67 expression varies across the cell cycle.
  • Consider using a nuclear counterstain or co-staining with cell cycle markers to confirm localization and specificity.
