Biological Background: IL-22 Function and Localization
- Interleukin-22 (IL-22, also known as IL-TIF, IL-22a) is a cytokine of the IL-10 family that plays a critical role in modulating tissue responses during inflammation and in the regeneration of epithelial cells to maintain barrier function after injury. Related references: PMID:33852830 PMID:35525330 PMID:33912578
- IL-22 signals through a heterodimeric receptor complex composed of IL22RA1 (specific receptor) and IL10RB (shared subunit), activating JAK1/TYK2 tyrosine kinases and downstream STAT3, ERK1/2, and PI3K/AKT pathways to promote cell survival and proliferation. Related references: PMID:24742671
- Downstream effects include phosphorylation of GSK3B at Ser-9 and CTTN, as well as promotion of epithelial cell spreading. Related references: PMID:24742671
- Unlike most cytokines, IL-22 does not directly target immune cells; its receptor IL22RA1 is primarily expressed on non-immune cells in various organs.
- The protein is secreted and contains a signal peptide, disulfide bonds, and N-linked glycosylation sites.
- A 3D structure is available, revealing a compact cytokine fold stabilized by disulfide bonds.
Experimental Guidance and Technical Tips
- This antibody was generated against a recombinant protein corresponding to amino acids 34-179 of mouse IL-22, encompassing the mature cytokine sequence. It is expected to recognize both native and recombinant mouse IL-22.
- The fluorophore is conjugated directly to the antibody, making it suitable for intracellular flow cytometry (FC intra). Ensure proper fixation and permeabilization protocols are used to detect cytoplasmic IL-22.
- Consider titrating the antibody to determine the optimal concentration for your specific sample and experimental conditions.
- Validate the antibody’s performance in a relevant mouse cell system known to express IL-22, such as activated Th17 or Th22 cells.
