Biological Background: Interleukin-22 Function and Localization
- Interleukin-22 (IL-22), also known as IL-10-related T-cell-derived-inducible factor (IL-TIF) or IL-TIF alpha, is a cytokine member of the IL-10 family.
- Plays a critical role in modulating tissue responses during inflammation and is essential for the regeneration of epithelial cells to maintain barrier function after injury. Related references: PMID:33852830, PMID:35525330, PMID:33912578
- Unlike most cytokines, IL-22 has no direct effect on immune cells. It signals through a heterodimeric receptor composed of IL22RA1 (present on non-immune cells) and IL10RB, activating JAK1/TYK2 and downstream STAT3, ERK1/2, and PI3K/AKT pathways. Promotes phosphorylation of GSK3B at Ser-9 and CTTN, and epithelial cell spreading. Related references: PMID:24742671
- Predominantly secreted; the protein contains a signal peptide and is found extracellularly.
- Post-translational modifications include disulfide bond formation and glycosylation, as suggested by database annotations.
Experimental Guidance and Technical Tips
- This antibody is supplied as an conjugate, suitable for direct intracellular flow cytometry. Optimize fixation and permeabilization protocols for your cell type.
- Titrate the antibody to determine the optimal staining concentration for your assay; include appropriate isotype controls.
- The immunogen comprises amino acids 34–179 of mouse IL-22, which may contribute to high specificity. Validation in other applications (e.g., Western blot) should be performed carefully.
- Consider using this reagent to study IL-22 expression in activated T cells or other lymphocyte subsets following appropriate stimulation.
