Biological Background: IL1R1/CD121a Function and Localization
- Interleukin-1 receptor type 1 (IL1R1), also known as CD121a, IL-1R alpha, p80, is encoded by the Il1r1 gene in mice.
- Functions as a receptor for interleukin-1 alpha (IL1A), interleukin-1 beta (IL1B), and the IL-1 receptor antagonist (IL1RN), binding these ligands with comparable affinity.
- Upon ligand binding, IL1R1 associates with the coreceptor IL1RAP to form a high-affinity signaling complex, leading to activation of NF-kappa-B and MAPK pathways.
- Signaling involves TIR domain-mediated recruitment of adaptor proteins such as TOLLIP, MYD88, IRAK1, or IRAK2.
- Isoform 2 is expressed in brain tissues and, together with IL1RAP isoform 3, mediates IL1B-induced neuronal activity, including potentiation of NMDA receptor-mediated calcium influx via Akt kinase activation.
- IL1R1 is a transmembrane glycoprotein with disulfide bonds and immunoglobulin-like domains, belonging to the interleukin-1 receptor family.
- Post-translational modifications include phosphorylation and glycosylation; a secreted form has also been identified.
- Plays a role in inflammatory responses and T-helper 1 (Th1) cell costimulation of IFNG production.

- The immunogen (amino acids 20–338) covers the extracellular domain of mouse IL1R1, making it suitable for detecting surface expression by flow cytometry without permeabilization.
- The conjugate is excited by the 488 nm laser and emits in the green channel (e.g., FITC/GFP); ensure compensation if used in multicolor panels.
- Titrate the antibody to determine the optimal concentration for your specific cell type and staining conditions; typical starting dilutions range from 1:50 to 1:200.
- Include an appropriate isotype control antibody conjugated to the same fluorophore to assess nonspecific binding.
