  • Interleukin-1 receptor type 1 (IL-1R1), also known as CD121a, is a receptor for IL-1A, IL-1B, and the natural antagonist IL-1RN.
  • Upon ligand binding, IL-1R1 associates with the co-receptor IL1RAP to form a high-affinity receptor complex that activates NF-kappa-B, MAPK, and other signaling pathways.
  • Signal transduction involves the recruitment of adapter molecules such as TOLLIP, MYD88, and IRAK1 or IRAK2 via their TIR domains, leading to downstream inflammatory responses.
  • Binding of the antagonist IL-1RN to IL-1R1 prevents the association with IL1RAP, thereby inhibiting the formation of a functional signaling complex.
