- Interleukin-1 receptor type 1 (IL-1R1), also known as CD121a, is a receptor for IL-1A, IL-1B, and the natural antagonist IL-1RN.
- Upon ligand binding, IL-1R1 associates with the co-receptor IL1RAP to form a high-affinity receptor complex that activates NF-kappa-B, MAPK, and other signaling pathways.
- Signal transduction involves the recruitment of adapter molecules such as TOLLIP, MYD88, and IRAK1 or IRAK2 via their TIR domains, leading to downstream inflammatory responses.
- Binding of the antagonist IL-1RN to IL-1R1 prevents the association with IL1RAP, thereby inhibiting the formation of a functional signaling complex.
- An alternative isoform (isoform 2) cannot mediate canonical IL-1 signaling but cooperates with IL1RAP isoform 3 to mediate IL-1B-induced neuronal activity, including NMDA receptor-potentiated calcium influx via Akt kinase.
- IL-1R1 is a transmembrane protein located on the cell membrane; it can also exist in a secreted form, likely through shedding.
- Isoform 2 is expressed in various brain tissues, indicating a specialized function in the central nervous system.

- The conjugate is suitable for direct flow cytometry (FC) applications; ensure appropriate compensation controls and laser/filter settings for optimal detection.
- Since the immunogen corresponds to the extracellular domain (amino acids 20–338), the antibody is expected to recognize the surface receptor; consider using a surface staining protocol without permeabilization for live-cell FC.
- IL-1R1 can be shed from the cell surface; validate surface expression under your experimental conditions, possibly using a positive control like activated macrophages or T cells.
- The antibody reacts specifically with mouse IL-1R1; cross-reactivity with other species is not expected. Validate in mouse-derived samples.
- For FC, titrate the antibody to determine the optimal staining concentration for your cell type.
