Biological Background: Interleukin-13 Function and Localization

  • Interleukin-13 (IL-13, also known as T-cell activation protein P600) is a 14 kDa secreted glycoprotein cytokine encoded by the Il13 gene in mouse.
  • IL-13 plays a central role in allergic inflammation and immune defense against parasite infection (PubMed:15361238). It synergizes with IL-2 to regulate interferon-gamma synthesis and stimulates proliferation of B cells and activation of eosinophils, basophils, and mast cells. Related references: PMID:15361238
  • It modulates IL-33 activity by regulating the production of transmembrane and soluble forms of interleukin-1 receptor-like 1 (IL1RL1) (PubMed:34789557). Related references: PMID:34789557
  • IL-13 antagonizes Th1-driven proinflammatory responses and downregulates synthesis of many proinflammatory cytokines including IL1, IL6, IL10, IL12, and TNF, partially via suppression of NF-kappa-B.
  • On nonhematopoietic cells, IL-13 induces vascular cell adhesion protein 1 (VCAM1) in endothelial cells, aiding eosinophil recruitment.
  • IL-13 signals through a receptor complex of IL4R and IL13RA1, activating JAK1, TYK2, and STAT6 (PubMed:34795444, PubMed:8871614). A high-affinity decoy receptor IL13RA2 mediates internalization and depletion of extracellular IL-13 (PubMed:29305434). Related references: PMID:34795444 PMID:8871614 PMID:29305434
  • In esophageal epithelium, IL-13 induces an inflammatory response that dysregulates desmosome intercellular adhesion junctions by downregulating desmoglein-1 (DSG1) (PubMed:24220297). Related references: PMID:24220297
  • IL-13 contains disulfide bonds and is glycosylated, which contribute to its structural stability and biological activity.

Experimental Guidance and Technical Tips

  • The immunogen is a recombinant fusion protein corresponding to amino acids 20-131 of mouse IL-13 (NP_032381.1), covering the mature secreted protein; this design should ensure reactivity with the native processed form.
  • This antibody is validated for intracellular flow cytometry (FC intra). Use standard permeabilization and fixation protocols suitable for cytokine detection.
  • Because IL-13 is secreted, consider adding a protein transport inhibitor (e.g., Brefeldin A or Monensin) during cell stimulation to accumulate intracellular cytokine for optimal detection.
  • As a directly conjugated monoclonal antibody (), it is ready-to-use for single-step staining. Verify signal in relevant mouse cell populations such as activated Th2 cells, mast cells, or basophils.
  • For multipanel flow cytometry, ensure spectral compatibility of with other fluorophores in your panel and perform appropriate compensation.
  • No cross-reactivity with other species has been reported; the antibody is specific for mouse IL-13. Validate performance in your experimental system.
