Biological Background: IgM Heavy Constant Region Function and Localization
- Immunoglobulin heavy constant mu (IgM heavy chain) is encoded by the Ighm gene in mice; it belongs to the immunoglobulin superfamily and represents the first antibody isotype expressed during B cell development.
- It functions as the constant region of both membrane-bound B cell receptors (BCR) and secreted immunoglobulin M (sIgM). The membrane form provides tonic survival signals and mediates pre-BCR signaling, while secreted IgM assembles into pentamers or hexamers for high-avidity antigen binding.
- Secreted IgM (sIgM) mediates humoral effector functions by recruiting complement component C1q to initiate the classical pathway and by binding high-affinity Fc receptor FCMR on lymphoid cells, promoting IgG responses while maintaining self-tolerance.
