Biological Background: IgM Function and Localization

  • IgM (Immunoglobulin heavy constant mu) is the heavy chain constant region of immunoglobulin M, existing as both membrane-bound B cell receptor (BCR) and secreted antibody (sIgM). The gene Ighm encodes several isoforms via alternative splicing.
  • Membrane-bound IgM forms the BCR complex, providing constitutive tonic signaling for B cell survival and mediating pre-BCR signaling that regulates B cell selection and allelic exclusion of Ig genes.
  • Secreted IgM multimerizes into mainly pentamers and hexamers, enabling multivalent antigen binding and high avidity. It mediates effector functions through Fc receptors (e.g., FCMR) and complement activation (C1q binding), promoting pathogen neutralization and clearance of apoptotic cells. • Natural sIgM is polyreactive, recognizing conserved self and pathogen structures, while immune sIgM is antigen-specific; both are essential for an efficient humoral immune response. Related references: PMID:10899913, PMID:28135254, PMID:34788614
  • IgM participates in mucosal immunity: it is transported across mucosal epithelium via PIGR, and IgM-antigen complexes undergo FCMR-mediated retrotranscytosis to mucosal lymphoid tissues. Related references: PMID:34788614
  • Subcellular localization: Secreted IgM is found in extracellular space and serum; membrane-bound IgM is anchored on the plasma membrane of B lymphocytes.
  • Tissue specificity: Expressed in bone marrow and at the protein level in B-1 and B-2 cells.
  • Key post-translational modifications include N-linked glycosylation and inter-chain disulfide bonds stabilizing the multimeric structure. The 3D structure of the Fc region has been resolved.

Experimental Guidance and Technical Tips

  • The immunogen spans amino acids 105-454 of mouse IgM (constant region), suggesting recognition of all IgM molecules irrespective of variable region. Validate specificity in your target cell system.
  • Directly conjugated to for flow cytometry (FC). Titrate to determine optimal staining concentration; use appropriate isotype controls and viability dyes.
  • For intracellular staining, consider permeabilization and validate conditions experimentally.
  • IgM is highly expressed on B cells; expect bright staining. Include Fc receptor blocking reagents if non-specific binding is observed.
