Biological Background: ICOS/CD278 Function and Localization
- Inducible T-cell costimulator (ICOS), also known as CD278, is a stimulatory receptor expressed on activated or antigen-experienced T-cells that plays a crucial role in the immune response. Upon binding to its ligand ICOSL on antigen-presenting cells, it enhances T-cell proliferation, cytokine secretion, and antibody production by B-cells. Related references: PMID:11343123, PMID:10657606
- ICOS belongs to the CD28 family and is also referred to as activation-inducible lymphocyte immunomediatory molecule (AILIM), CD28 and CTLA-4-like protein (CCLP), and CD28-related protein 1 (CRP-1). It is essential for efficient T-cell/B-cell interaction and for CD40-mediated class switching of immunoglobulin isotypes. Related references: PMID:11343122
- Mechanistically, ICOS potentiates TCR-induced calcium flux by augmenting PLCG1 activation and actin remodeling, and independently activates PI3K signaling pathways. Related references: PMID:27693916, PMID:19915142
- It is critical for the differentiation of T follicular regulatory cells during immune challenges such as viral infection. Related references: PMID:36754569
- ICOS is a single-pass type I transmembrane glycoprotein containing an immunoglobulin domain in its extracellular region, a disulfide bond, and a signal peptide.
- Its tissue distribution includes activated T-cells, resting memory T-cells, thymic medulla, germinal centers, T-cell zones of lymph nodes, and Peyer's patches, with low expression in the spleen.
- The protein has a predicted molecular weight of approximately 23 kDa (Swiss-Prot: 22709 Da), consistent with the observed weight of ~22 KDa for this antibody.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (amino acids 21–142) of mouse ICOS, which is exposed on the cell surface, suggesting suitability for live-cell staining in flow cytometry.
- For flow cytometry, consider titrating the antibody to determine the optimal concentration for your specific cell type and experimental conditions.
- Include appropriate isotype controls to account for non-specific binding.
- If fixation and permeabilization are required for intracellular staining, validate antibody compatibility, as the epitope may be affected.
- Pre-incubation with Fc blocking reagents may reduce background staining in samples with high Fc receptor expression.
