Biological Background: ICOS/CD278 Function and Localization
- Inducible T-cell costimulator (ICOS), also known as CD278, is a stimulatory receptor expressed on activated T-cells and resting memory T-cells. It is highly expressed in the thymic medulla, germinal centers, and T-cell zones of lymph nodes and Peyer's patches, with lower levels in the spleen.
- Upon binding to its ligand ICOSL on antigen-presenting cells, ICOS delivers costimulatory signals that enhance T-cell proliferation, lymphokine secretion (e.g., IL-10), upregulation of cell-cell interaction molecules, and effective help for antibody secretion by B-cells. Related references: PMID:10657606
- Acts as a costimulatory receptor critical for the differentiation of T follicular regulatory cells during immune challenges such as viral infection. Related references: PMID:36754569
- Mechanistically, potentiates TCR-induced calcium flux by augmenting PLCG1 activation and actin remodeling, and activates PI3K signaling pathways independently of calcium flux. Related references: PMID:27693916, PMID:19915142
- Prevents the apoptosis of pre-activated T-cells.
- Essential for efficient interaction between T and B-cells and for normal antibody responses to T-cell-dependent antigens.
- Plays a critical role in CD40-mediated class switching of immunoglobulin isotypes. Related references: PMID:11343122
- As a type I transmembrane protein, ICOS localizes to the cell membrane and contains an immunoglobulin-like domain, a signal peptide, and conserved disulfide bonds that contribute to its structure and function.
Experimental Guidance and Technical Tips
- The immunogen used is a recombinant fusion protein corresponding to amino acids 21–142 of mouse ICOS/CD278 (NP_059508.2), which spans the extracellular domain; this design favors generation of antibodies recognizing native surface-expressed ICOS in flow cytometry.
- This conjugate is optimized for direct flow cytometry; consider titrating the antibody to determine the optimal staining concentration for your specific cell model and instrument settings.
- (equivalent to Alexa Fluor® 647) is compatible with standard red laser (633–647 nm) excitation and 660/20 nm bandpass filters, allowing multiplexing with other fluorophore-conjugated antibodies.
- Since ICOS is upregulated upon T-cell activation, include appropriate positive and negative controls (e.g., resting vs. activated T cells) in your experiments to confirm specificity.
- Perform Fc receptor blocking and use appropriate gating strategies to minimize non-specific binding and ensure accurate identification of ICOS-positive populations.
