Biological Background: FOXP3 Function and Localization
- FOXP3 is a transcriptional regulator essential for the development and suppressive function of regulatory T-cells (Treg), critical for immune homeostasis. Related references: PMID:22813742
- It can act as either a transcriptional repressor or activator, coordinating with histone acetylases and deacetylases to modulate gene expression.
- FOXP3 suppresses cytokines by repressing IL2 and IFNG expression through interactions with transcription factors like RELA, NFATC2, RUNX1, and IKZF4. Related references: PMID:15790681 PMID:17377532 PMID:19696312
- It also activates Treg-associated genes like CTLA4, TNFRSF18, and IL2RA, partly via RUNX1. Related references: PMID:17377532
- FOXP3 inhibits Th17 cell differentiation by antagonizing RORC, reducing IL17 production. Related references: PMID:18368049
- Subcellularly, it is found in both the nucleus and cytoplasm.
- Foxp3 shows high expression in thymus and spleen, consistent with its role in immune regulation.
Experimental Guidance and Technical Tips
- This antibody is raised against a synthetic peptide corresponding to the N-terminal region (amino acids 1-100) of mouse Foxp3.
- For intracellular flow cytometry (FC intra), consider appropriate fixation and permeabilization protocols to enable access to the nuclear/cytoplasmic FOXP3 protein.
- Titrate the antibody to determine the optimal working concentration for your specific assay and cell type.
- Validate staining specificity in known Treg populations (e.g., CD4+CD25+ cells) or Foxp3-expressing cell lines.
- Cross-reactivity with human or other species has not been characterized; use on mouse samples is supported.
