Biological Background: Flt3/CD135 Function and Localization
- Receptor-type tyrosine-protein kinase FLT3 (also known as CD135, FLK-2, fetal liver kinase 2) acts as a cell-surface receptor for the cytokine FLT3LG, regulating differentiation, proliferation and survival of hematopoietic progenitor cells and dendritic cells.
- Upon ligand binding, FLT3 promotes phosphorylation of SHC1, AKT1 and activation of MTOR, as well as RAS-dependent signaling via MAPK1/ERK2 and MAPK3/ERK1. It also mediates phosphorylation of FES, FER, PTPN6/SHP, PTPN11/SHP-2, PLCG1, and STAT5A/STAT5B, though wild-type FLT3 activation of STAT5 is marginal.
- Mutations causing constitutive kinase activity drive cell proliferation and apoptosis resistance through multiple downstream pathways, highlighting its proto-oncogenic role.
- Tissue expression is predominantly in hematopoietic stem and progenitor cell-enriched populations, with additional detection in brain, placenta and testis.
- Subcellularly, FLT3 localizes to the plasma membrane and the endoplasmic reticulum lumen.
- The protein features extracellular immunoglobulin domains, a single transmembrane helix, and an intracellular tyrosine kinase domain. Key post-translational modifications include glycosylation, disulfide bond formation, and ligand-induced autophosphorylation.
- Keywords associated with FLT3: ATP-binding, Disulfide bond, Glycoprotein, Immunoglobulin domain, Kinase, Membrane, Nucleotide-binding, Phosphoprotein, Proto-oncogene, Receptor, Transferase, Transmembrane helix, Tyrosine-protein kinase, Ubl conjugation.
Experimental Guidance and Technical Tips
- The immunogen comprises a recombinant fusion protein spanning amino acids 28–544 of mouse CD135 (NP_034359.2). Consider verifying reactivity with the native protein on primary mouse hematopoietic cells.
- For flow cytometry, titrate the antibody on mouse spleen or bone marrow cells to determine the optimal staining concentration. Use Fc receptor blocking to minimize non-specific binding.
- When designing multicolor panels, account for the fluorophore’s spectral properties and perform appropriate compensation controls.
- The antibody is recommended for mouse samples; cross-reactivity with other species has not been reported and should be tested independently if required.
- Validate specificity by comparing staining in Flt3-knockout models or with cell lines differentially expressing CD135.
- As FLT3 functions in hematopoietic stem cell maintenance and dendritic cell development, this reagent can support immunophenotyping, leukemia stem cell detection, or investigation of FLT3-ITD-driven signaling.
