Biological Background: CD8a Function and Localization
- T-cell surface glycoprotein CD8 alpha chain (CD8a), also known as Lyt-2, is an integral membrane glycoprotein essential for adaptive immunity.
- Functions as a coreceptor for MHC class I molecules, interacting simultaneously with the T-cell receptor (TCR) and MHC I on antigen-presenting cells.
- Recruits the Src kinase LCK to the TCR-CD3 complex upon antigen recognition, initiating intracellular signaling pathways leading to T-cell activation and lymphokine production.
- Enables cytotoxic T-lymphocytes (CTLs) to recognize and eliminate infected or tumor cells through the MHC class I pathway.
- In NK cells, CD8A homodimers promote survival, conjugation, and lysis of multiple target cells, and support differentiation into memory CD8+ T-cells.
- Localized to the cell membrane, with structural features including a signal peptide, transmembrane helix, and disulfide bonds; also undergoes glycosylation and palmitoylation.
- Belongs to the immunoglobulin superfamily and is subject to alternative splicing, generating isoforms that fine-tune immune responses.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (amino acids 28–196) of mouse CD8a, suggesting the antibody recognizes surface-expressed CD8a; confirm expression in your cell system.
- is a red fluorescent dye suitable for flow cytometry; optimize antibody titration and include appropriate isotype controls to minimize non-specific binding.
- For staining mouse splenocytes or lymphocyte preparations, consider Fc receptor blocking to reduce background.
- Validate staining specificity using CD8a knockout models or competitive blocking with unconjugated antibody if available.
