Biological Background: C5aR1 Function and Localization
- C5aR1 (CD88) is a G protein-coupled receptor for the potent anaphylatoxin C5a, a key mediator of inflammatory and immune responses.
- Ligand binding stimulates chemotaxis, granule enzyme release, intracellular calcium mobilization, and superoxide anion production via G(i)/G(o) protein-mediated inhibition of adenylate cyclase. Related references: PMID:37852260
- The receptor is predominantly localized at the cell membrane and can also be found in cytoplasmic vesicles.
- C5aR1 contains seven transmembrane helices, conserved disulfide bonds, and undergoes post-translational modifications such as glycosylation, phosphorylation, and sulfation.
- The extracellular N-terminus provides a high-affinity binding site for C5a, while the transmembrane region is responsible for initiating downstream signaling.
Experimental Guidance and Technical Tips
- This conjugate is suitable for direct flow cytometry (FC) and targets the N-terminal extracellular domain (amino acids 1-37) of mouse C5aR1.
- For optimal staining, validate the antibody in the relevant mouse cell or tissue samples and include appropriate isotype controls and viability dyes.
- Given the GPCR nature of the target, consider assessing receptor surface expression under non-permeabilizing conditions and verify specificity using C5aR1 knockout or knockdown controls.
- As a monoclonal rabbit antibody, it typically offers high affinity and reproducibility; however, titration is recommended to determine the optimal working concentration for your assay.
