  • CD82, also known as C33, IA4, Kai1, or Tspan27, is a member of the tetraspanin superfamily and functions as a structural component of tetraspanin-enriched microdomains (TERMs) at the plasma membrane.
  • It plays a multifaceted role in regulating cell signal transduction, adhesion, migration, and protein trafficking, particularly by attenuating epidermal growth factor (EGF) signaling through ligand-induced EGFR endocytosis and ubiquitination.
  • CD82 enhances cell-matrix adhesion by organizing integrin alpha4/ITA4 at the membrane and suppresses migration mediated by integrins alpha6/ITGA6 and beta1/ITGB1.
  • In immune cells, CD82 associates with CD4 or CD8 to deliver costimulatory signals for the TCR/CD3 pathway, restrains phagocyte migration, and supports macrophage activation, including promoting anti-inflammatory phenotypes. Related references: PMID:35754722
