Biological Background: CD69 Function and Localization
- CD69 (Early activation antigen CD69), also known as AIM and VEA, is a transmembrane protein belonging to the C-type lectin family and serves as an early activation marker on immune cells.
- It is rapidly upregulated on the surface of platelets, T-lymphocytes, and NK cells upon activation by antigen recognition or cytokine signaling, playing a critical role in immune cell homeostasis (PubMed:20696842, PubMed:38540778). Related references: PMID:20696842, PMID:38540778
- CD69 negatively regulates Th17 cell differentiation through carbohydrate-dependent interaction with galectin-1 on immature dendritic cells and via JAK3/STAT5 signaling that controls RORγt transcription (PubMed:20696842). Related references: PMID:20696842
- The protein also promotes regulatory T-cell conversion by engaging the S100A8/S100A9 complex and acts as an oxidized low-density lipoprotein (oxLDL) receptor in CD4 T-cells, negatively regulating inflammation by inducing PDCD1 expression through NFAT activation.

Experimental Guidance and Technical Tips
- The immunogen comprises amino acids 64–199 of mouse CD69, corresponding to the extracellular C-type lectin domain, making the antibody suitable for detecting cell surface expression on live, unfixed cells by flow cytometry.
- For flow cytometry, titrate the antibody to determine the optimal working concentration. Include appropriate isotype controls and single-stain compensation controls when using multiple fluorophores.
- Validate antibody performance using known positive controls (e.g., activated mouse T-cells or NK cells) and confirm specificity with CD69-deficient cells or blocking peptides if available.
- The conjugate is compatible with standard red/orange laser lines; ensure instrument settings are appropriately configured to avoid spillover.
