Biological Background: CEACAM1 Function and Localization
- CEACAM1, also known as CD66a and BGP-1, is encoded by Ceacam1 and exists as multiple isoforms (30–57 kDa) generated by alternative splicing.
- Mediates calcium-independent homophilic cell adhesion, contributing to cell-cell interactions. Related references: PMID:1633107
- Functions as a coinhibitory receptor in immune cells; upon engagement, recruits PTPN6 to inhibit TCR signaling in T cells, KLRK1-mediated cytotoxicity in NK cells, and CSF3R/IL1B pathways in neutrophils. Related references: PMID:17081782, PMID:21029969, PMID:22496641, PMID:23696226
- Serves as a receptor for murine hepatitis virus (MHV), facilitating viral entry via the spike glycoprotein.
- Plays pro-angiogenic roles in endothelial cell differentiation, blood vessel remodeling, and vascular permeability regulation through VEGFR2 signaling. Related references: PMID:16680193, PMID:22962327, PMID:21081647
- Shows broad tissue expression, high in liver, femur, and immune cells (granulocytes, lymphocytes, neutrophils, monocytes/macrophages); intestinal T-cells preferentially express isoform 2. Related references: PMID:11994468, PMID:25490771, PMID:21029969, PMID:23123061, PMID:25908210
- Localizes to lateral, apical, and basal plasma membranes, adherens junctions, microvilli, and cytoplasmic vesicles.
- Undergoes phosphorylation by kinases such as LCK and INSR, enabling interactions with PTPN6 and SHC1 to modulate downstream signaling pathways. Related references: PMID:22496641, PMID:18544705, PMID:15467833
Experimental Guidance and Technical Tips
- The immunogen (aa 35-428) spans the extracellular domain, making this antibody suitable for detecting surface-expressed CEACAM1 by flow cytometry.
- Consider using appropriate isotype controls and validate staining in relevant mouse immune cell populations (e.g., splenocytes, PBMCs) or CEACAM1-expressing cell lines.
- Due to multiple isoforms, confirm antibody reactivity in your specific sample type; optimal working dilution should be determined empirically.
