Biological Background: CD6 Function and Localization
- CD6 is a single-pass type I transmembrane glycoprotein belonging to the scavenger receptor cysteine-rich (SRCR) superfamily, with multiple isoforms generated by alternative splicing (observed at 64, 68, and 72 kDa).
- It functions as a cell adhesion molecule that mediates T-cell interactions with antigen-presenting cells via binding to ALCAM/CD166.
- Contributes to signaling cascades downstream of the TCR/CD3 complex, acting as a costimulatory receptor that enhances T-cell activation and proliferation. Related references: PMID:24584089
- Promotes the formation and maturation of the immunological synapse, facilitating stable T-cell–APC contacts.
- Acts as a calcium-dependent pattern recognition receptor, binding both Gram-positive and Gram-negative bacteria (LPS and lipoteichoic acid), which triggers MAP kinase activation and pro-inflammatory cytokine secretion.
- Localizes to the cell membrane and is predominantly expressed in lymphoid tissues, including thymus, lymph node, and spleen.
- Post-translational modifications include N-linked glycosylation, disulfide bond formation, and phosphorylation on cytoplasmic tyrosine residues.
Experimental Guidance and Technical Tips
- For flow cytometry, titrate the antibody and include an appropriate isotype control to determine optimal staining concentrations and minimize background.
- The immunogen (amino acids 20–155 in the extracellular domain) is expected to generate antibodies that recognize native cell surface CD6; confirm performance in your assay system.
- Validate staining in mouse lymphoid tissues (thymus, lymph node, spleen) where CD6 expression is highest; co-staining with ALCAM/CD166 may be used to study CD6-mediated adhesion.
