Biological Background: CD5 Function and Localization
- CD5 is a lymphoid-specific receptor expressed on all T-cells and the B1a subset of B-cells, playing a role in TCR/BCR signaling regulation, thymocyte selection, and immune tolerance. Related references: PMID:35720357

- CD5 functions as a negative regulator of TCR signaling during thymocyte development by recruiting signaling proteins such as LCK, CD3ζ, PI3K, and CBL. Related references: PMID:11157848

- Use appropriate isotype-matched control antibodies and include viability dyes to discriminate live/dead cells and minimize background.
- Protect the antibody from light during staining and storage, as repeated exposure may reduce fluorescence.
- Standard staining protocols recommend incubating 10⁶ cells in 100 µL of staining buffer for 20–30 minutes on ice in the dark, followed by washing.
