Biological Background: CD5 Function and Localization
- CD5, also known as Ly-1 or Lyt-1, is a lymphoid-specific cell surface receptor expressed on all T-cells and in a subset of B1a cells.
- Plays a critical role in regulating T-cell receptor (TCR) and B-cell receptor (BCR) signaling, influencing thymocyte selection, T-cell effector differentiation, and immune tolerance. Related references: PMID:35720357
- Mediates contact-dependent T-B cell interactions by binding to ligands on B-cells, such as CD5L or CD72, thereby promoting T-dependent B-cell activation and maintaining regulatory T and B-cell homeostasis. Related references: PMID:24950378 | PMID:9064341 | PMID:9723705
- Acts as a negative regulator of TCR signaling during thymocyte development; associates with signaling proteins including LCK, CD3ζ chain, PI3K, and CBL, and its co-engagement with CD3 enhances CBL recruitment, leading to VAV1 phosphorylation and degradation (By similarity). Related references: PMID:11157848
- Modulates B-cell biology through ERK1/2 activation in a Ca²⁺-dependent pathway via the non-selective Ca²⁺ channel TRPC1, leading to IL-10 production (By similarity).
- Localizes to the cell membrane and is classified as a glycoprotein, phosphoprotein, and disulfide-bonded receptor with signal and transmembrane domains, consistent with its 54 kDa molecular weight.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular region (aa 25-371) of mouse CD5; the antibody may recognize an epitope within this domain, making it suitable for detecting surface CD5 in flow cytometry.
- Since it is directly conjugated with (equivalent to Alexa Fluor 594), no secondary antibody is required; titrate the antibody to determine the optimal working concentration for your cell samples.
- Validate staining in appropriate mouse tissue or cell lines (e.g., mouse splenocytes, thymocytes, or B1a cells) using an isotype control to assess specificity.
- Store at 2-8°C protected from light and avoid repeated freeze-thaw cycles to maintain conjugate stability.
