Biological Background: CD5 Function and Localization
-
CD5 is a lymphoid-specific receptor expressed on all T-cells and a subset of B-cells (B1a cells), also known as Ly-1.
-
Functions as a negative regulator of TCR signaling during thymocyte development by interacting with signaling proteins such as LCK, CD3ζ, PI3K, and CBL. Related references: PMID:11157848
-
Involved in T-dependent B-cell activation through interaction with CD5L and CD72, contributing to regulatory T and B-cell homeostasis. Related references: PMID:24950378 PMID:9064341 PMID:9723705
-
Modulates B-cell biology via ERK1/2 activation in a Ca2+-dependent pathway through TRPC1, leading to IL-10 production.
-
Localizes to the cell membrane as a single-pass type I transmembrane protein.
-
Contains a signal peptide, three scavenger receptor cysteine-rich (SRCR) repeats in the extracellular domain, and a cytoplasmic tail; it is glycosylated and phosphorylated.
-
Regulates thymocyte selection, T-cell effector differentiation, and immune tolerance. Related references: PMID:35720357
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular region (aa 25-371), so the antibody likely recognizes a cell surface epitope, making it well-suited for flow cytometry (FC) without permeabilization.
- For mouse samples, validate expression on T-cells (positive) and B1a B-cells; may also detect low levels on other lymphocyte subsets.
- Use appropriate isotype controls and Fc receptor blocking to minimize non-specific binding.
- Optimize antibody concentration by titration for each cell type.
- Consider co-staining with other T-cell markers (e.g., CD3, CD4, CD8) to characterize T-cell subsets.
- As a monoclonal rabbit antibody, it may offer high specificity and affinity.
