Biological Background: CD48 Antigen Function and Localization
- CD48 antigen (SLAMF2) is a glycosylphosphatidylinositol (GPI)-anchored cell surface glycoprotein and a member of the signaling lymphocytic activation molecule (SLAM) family. Related references: PMID:21278219 PMID:9881969
- It interacts via its N-terminal immunoglobulin domain with receptors such as CD244/2B4 and CD2, regulating immune cell activation and function in T-lymphocytes and NK-cells. Related references: PMID:21278219 PMID:9881969
- CD48 participates in T-cell signaling by associating with CD2, which facilitates the recruitment of the Src family kinase LCK and the adaptor LAT to the TCR/CD3 complex, promoting LCK phosphorylation and downstream activation. Related references: PMID:9881969
- It induces phosphorylation of the cytoplasmic immunoreceptor tyrosine-based switch motifs (ITSMs) of CD244, initiating signaling cascades that lead to immunological synapse formation and directed release of cytolytic granules containing perforin and granzymes by T-lymphocytes and NK-cells. Related references: PMID:21278219 PMID:9881969
- Subcellular localization includes the cell membrane and membrane rafts; a secreted form has also been identified.
- Key structural features include a signal peptide, an extracellular region with one Ig-like V-type domain, a GPI-anchor, and two disulfide bonds.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (amino acids 23–216) of mouse CD48, covering the region involved in ligand binding, which may favor detection of native surface-expressed antigen.
- As an -conjugated primary antibody, it is ready-to-use for flow cytometry; however, titration is recommended to determine the optimal staining concentration for each experimental system.
- Consider including appropriate isotype controls and viability dyes to exclude non-specific binding and dead cells when analyzing CD48 expression on lymphocyte populations.
- Because CD48 is expressed on various immune cells, validate target cell identity with lineage markers in multicolor panels.
