Biological Background: CD48 Function and Localization

  • CD48 is a glycosylphosphatidylinositol (GPI)-anchored cell surface glycoprotein belonging to the SLAM family (SLAMF2). It interacts via its N-terminal immunoglobulin domain with receptors CD244 (2B4) and CD2 to regulate immune cell activation. Related references: PMID:21278219 PMID:9881969
  • In T-cells, CD48 associates with CD2, facilitating the recruitment of the kinase LCK and adaptor LAT to the TCR/CD3 complex, enhancing LCK phosphorylation and downstream signaling. Related references: PMID:9881969
  • CD48 engagement triggers phosphorylation of immunoreceptor tyrosine-based switch motifs (ITSMs) on CD244, leading to formation of immunological synapses and directed release of cytolytic granules by T-lymphocytes and NK-cells. Related references: PMID:21278219 PMID:9881969
  • CD48 is localized primarily at the cell membrane and within membrane rafts; a soluble secreted form also exists.
  • Keywords include: GPI-anchor, glycoprotein, immunoglobulin domain, disulfide bond, and signal peptide, indicating post-translational modifications important for its function.
  • The protein is also known as BCM1, BLAST-1, HM48-1, MRC OX-45, and sgp-60, reflecting its discovery history.

Experimental Guidance and Technical Tips

  • This antibody was generated against the extracellular domain (aa 23–216) of mouse CD48 and is expected to recognize the native protein on live cells by flow cytometry.
  • For flow cytometry, titrate the antibody to determine optimal staining concentration. Use an appropriate isotype control and ensure single-cell suspension.
  • Validate staining on known CD48-expressing mouse immune cell subsets (e.g., T cells, NK cells) and use a viability dye to exclude dead cells.
  • Since CD48 is a GPI-anchored protein, fixation/permeabilization protocols should be evaluated for their effect on epitope accessibility.
