Biological Background: CD48 Function and Localization
- Mouse CD48, also known as BCM1, BLAST-1, SLAMF2, or sgp-60, is a member of the signaling lymphocyte activation molecule (SLAM) family of immunoglobulin-like receptors.
- CD48 serves as a high-affinity ligand for CD2 and 2B4 (CD244), playing a pivotal role in modulating innate and adaptive immune responses. Related references: PMID:21278219 PMID:9881969
- Upon binding to CD2, CD48 facilitates the recruitment of the Src family kinase LCK and the adaptor protein LAT to the T-cell receptor (TCR)/CD3 complex, thereby enhancing LCK phosphorylation and downstream signaling. Related references: PMID:9881969
- CD48 engagement of 2B4 (CD244) triggers phosphorylation of immunoreceptor tyrosine-based switch motifs (ITSMs) in the cytoplasmic tail of CD244, leading to formation of the immunological synapse and directed release of perforin- and granzyme-containing lytic granules from cytotoxic T lymphocytes and natural killer (NK) cells. - CD48 is a glycosylphosphatidylinositol (GPI)-anchored protein predominantly found in lipid raft microdomains of the plasma membrane; a soluble ectodomain form is also released by proteolytic shedding or phospholipase cleavage.
- The mature protein contains an N-terminal immunoglobulin V-type domain, an O-glycosylated mucin-like stalk, and a C-terminal GPI anchor; its calculated molecular weight is approximately 27.4 kDa.
