Biological Background: CD47 Function and Localization
- Leukocyte surface antigen CD47, also known as Integrin-associated protein (IAP), is encoded by the Cd47 gene.
- Serves as an adhesive protein and receptor for thrombospondin THBS1, modulating integrin signaling via heterotrimeric G proteins; involved in signal transduction, cardiovascular homeostasis, inflammation, apoptosis, angiogenesis, cellular self-renewal, and immunoregulation. Related references: PMID:20610415, PMID:23591719, PMID:27742621
- Plays a role in memory formation and synaptic plasticity in the hippocampus (By similarity).
- Acts as a receptor for SIRPA, preventing dendritic cell maturation and inhibiting cytokine production; interaction with SIRPG mediates cell-cell adhesion and costimulates T-cell activation (By similarity).
- Positively modulates FAS-dependent apoptosis in T-cells, perhaps by enhancing FAS clustering. Related references: PMID:15917238
- Suppresses angiogenesis, negatively modulates wound healing, inhibits stem cell self-renewal (via POU5F1, SOX2, MYC, KLF4), and may contribute to metabolic dysregulation during aging. Related references: PMID:32679764, PMID:18156939, PMID:23591719
- Prevents premature elimination of red blood cells. Related references: PMID:10856220
- Cell membrane localization; contains signal peptide, immunoglobulin domain, disulfide bond, transmembrane helix; undergoes glycosylation and phosphorylation; subject to alternative splicing.
Experimental Guidance and Technical Tips
- Since this antibody is directly conjugated to , it is ready for flow cytometry without a secondary detection reagent.
- The immunogen (aa 19-135) corresponds to the extracellular N-terminal region of mouse CD47, which includes the immunoglobulin-like domain; this design is expected to recognize cell surface CD47.
- For FC, consider performing antibody titration to determine the optimal concentration for your specific cell type.
- Validate the staining pattern in a relevant mouse cell line or primary cells known to express CD47 (e.g., macrophages, T-cells) and include appropriate isotype controls.
- Use Fc receptor blocking reagents to minimize non-specific binding if working with Fc receptor-rich samples.
