Biological Background: CD47 Function and Localization
- Leukocyte surface antigen CD47 (integrin-associated protein IAP, encoded by Cd47) is an adhesive protein that mediates cell-cell interactions and acts as a receptor for thrombospondin THBS1. Alternative splicing produces multiple isoforms.
- It is a multi-transmembrane immunoglobulin superfamily member localized to the cell membrane, undergoing post-translational modifications including glycosylation, disulfide bond formation, and phosphorylation.
- CD47 plays broad roles in signal transduction, cardiovascular homeostasis, inflammation, apoptosis, angiogenesis, cellular self-renewal, and immunoregulation. Related references: PMID:20610415 PMID:23591719 PMID:27742621
- In response to THBS1, CD47 modulates nitrous oxide signaling, acting as a pressor agent to support blood pressure. Related references: PMID:20610415
- CD47 positively regulates FAS-dependent apoptosis in T-cells, possibly by enhancing FAS clustering. Related references: PMID:15917238
- THBS1 engagement inhibits stem cell self-renewal via CD47, likely through regulation of key stem cell transcription factors. Related references: PMID:23591719
- CD47 suppresses angiogenesis and may be involved in metabolic dysregulation during normal aging. Related references: PMID:32679764
- In THBS1-dependent manner, CD47 negatively modulates wound healing and may prevent premature elimination of red blood cells. Related references: PMID:18156939 PMID:10856220
Experimental Guidance and Technical Tips
- The immunogen covers the extracellular domain (amino acids 19-135) of mouse CD47, suggesting the antibody targets the N-terminal region involved in ligand binding and cell-surface recognition.
- For flow cytometry, optimize antibody concentration by titration on mouse cell lines or primary cells known to express CD47; include appropriate isotype controls to ensure specificity.
- Ensure proper sample preparation (single-cell suspension and Fc receptor blocking) to minimize nonspecific binding, especially when analyzing mouse leukocytes.
- Use directly labeled conjugate to avoid secondary antibody steps, reducing background and simplifying the staining protocol.
