- CD45, encoded by the Ptprc gene, is a receptor-type tyrosine-protein phosphatase (PTPase) also known as leukocyte common antigen (L-CA), Ly-5, and T200. It is expressed on all nucleated hematopoietic cells.
- The protein is essential for T-cell activation through the antigen receptor, acting as a positive regulator of coactivation upon binding to DPP4.
- CD45 contains two intracellular PTPase domains: the first has catalytic activity, while the second modulates substrate specificity.
- It dephosphorylates key signaling molecules such as SKAP1, FYN, and LYN, thereby regulating T-cell receptor signaling thresholds.
