Biological Background: CD44 Antigen Function and Localization

  • CD44 antigen (also known as Ly-24, Pgp-1, HERMES, ECMR-III, hyaluronate receptor) is a multifunctional cell-surface receptor encoded by the Cd44 gene in mice. It plays critical roles in cell-cell interactions, adhesion, and migration, enabling cells to sense and respond to the tissue microenvironment.
  • CD44 serves as a receptor for hyaluronic acid (HA) and other extracellular matrix components such as collagen, growth factors, cytokines, or proteases, and participates in lymphocyte homing, T-cell activation, hematopoiesis, inflammation, and response to bacterial infection. Related references: PMID:25065622, PMID:8343954
  • Through its cytoplasmic domain, CD44 assembles signaling complexes involving PKN2, RhoGTPases (RAC1, RHOA), Rho-kinases, and phospholipase C, which regulate cytoskeletal reorganization and calcium mobilization required for cell motility and adhesion.
  • Upon binding to galectin-9 (LGALS9), CD44 activates LCK, ERK, and MAPK pathways, promoting natural killer (NK) cell activation. Related references: PMID:37006235
  • CD44 localizes to the cell membrane, particularly enriched in microvilli, and can also be secreted in a soluble form.
  • Tissue expression includes the intestinal epithelium.
  • Key post-translational modifications include glycosylation, phosphorylation, sulfation, and proteoglycan attachment; the protein contains disulfide bonds and a transmembrane helix.

Experimental Guidance and Technical Tips

  • This antibody is provided as an conjugate, suitable for direct flow cytometry (FC) without a secondary antibody. Validate in appropriate mouse samples or cell lines expressing CD44.
  • The immunogen corresponds to the extracellular domain (amino acids 23-176), which is expected to recognize the native CD44 antigen; consider testing on live cells.
  • For FC, titrate the antibody to determine the optimal concentration for your specific experimental conditions.
  • Note that CD44 is expressed on many cell types, including lymphocytes and epithelial cells; ensure appropriate gating strategies to analyze cell subsets.
  • As a rabbit monoclonal antibody, it offers high batch-to-batch consistency; validate specificity using CD44-negative cells or isotype controls.
  • The predicted protein molecular weight is 85 kDa, but CD44 often runs at higher apparent molecular weights due to extensive glycosylation. This antibody is intended for flow cytometry, not Western blotting; confirm performance under native conditions.
