Biological Background: CD4 Function and Localization

  • CD4, also known as L3T4 or Ly-4, is a T-cell surface glycoprotein that serves as a coreceptor for MHC class II molecules, playing a critical role in T-cell activation and immune response.
  • It interacts simultaneously with the T-cell receptor (TCR) and MHC class II on antigen-presenting cells, recruiting the Src kinase LCK to initiate intracellular signaling cascades leading to lymphokine production, adhesion, and T-helper cell activation.
  • CD4 is a member of the immunoglobulin superfamily, containing four extracellular Ig-like domains, a single transmembrane helix, and a short cytoplasmic tail.
  • Beyond T-cells, CD4 is also expressed on other immune cells such as macrophages, dendritic cells, and NK cells, where it participates in differentiation, cytokine expression, and cell migration through TCR-independent pathways.
  • The protein localizes to the cell membrane and is subject to multiple post-translational modifications including glycosylation, palmitoylation, and phosphorylation, which may regulate its function and membrane association.
  • CD4 is highly expressed in T-helper cells and is essential for thymic development of T-helper cells; it also triggers monocyte differentiation into mature macrophages.
  • Alternative splicing of the Cd4 gene results in multiple transcript variants, though the functional significance of these isoforms in the mouse is not fully characterized.

Experimental Guidance and Technical Tips

  • The immunogen spans amino acids 27–394 of mouse CD4, covering most of the extracellular region, so this antibody is expected to recognize the native, surface-expressed protein in flow cytometry.
  • For flow cytometry, titrate the antibody to identify the optimal concentration for your cell type and experimental conditions, and include appropriate isotype control and compensation controls.
  • This antibody is conjugated to , a red fluorescent dye; ensure your flow cytometer has the appropriate laser and filter setup (excitation ~590 nm, emission ~617 nm) and check for spectral overlap.
