Biological Background: CD4 Function and Localization
- T-cell surface glycoprotein CD4, also known as L3T4 or Ly-4, is an integral membrane glycoprotein essential for immune responses.
- Functions as a coreceptor for MHC class II molecules during T-cell activation. Interacts with the T-cell receptor (TCR) and MHC class II:peptide complexes on antigen-presenting cells, and recruits the Src kinase LCK to initiate intracellular signaling cascades leading to T-helper cell activation, cytokine production, and proliferation.
- Participates in the development of T-helper cells in the thymus and triggers the differentiation of monocytes into functional mature macrophages.
- Subcellular localization: Cell membrane; single-pass type I membrane protein.
- Highly expressed in T-helper cells, but also present in other immune cells such as macrophages, dendritic cells, and NK cells.
- Post-translational modifications include glycosylation, palmitoylation, phosphorylation, and disulfide bond formation.
- The mouse Cd4 gene undergoes alternative splicing, producing isoforms that may influence function and expression.
