Biological Background: CD4 Function and Localization

  • T-cell surface glycoprotein CD4, also known as L3T4 or Ly-4, is an integral membrane glycoprotein essential for immune responses.
  • Functions as a coreceptor for MHC class II molecules during T-cell activation. Interacts with the T-cell receptor (TCR) and MHC class II:peptide complexes on antigen-presenting cells, and recruits the Src kinase LCK to initiate intracellular signaling cascades leading to T-helper cell activation, cytokine production, and proliferation.
  • Participates in the development of T-helper cells in the thymus and triggers the differentiation of monocytes into functional mature macrophages.
  • Subcellular localization: Cell membrane; single-pass type I membrane protein.
  • Highly expressed in T-helper cells, but also present in other immune cells such as macrophages, dendritic cells, and NK cells.
  • Post-translational modifications include glycosylation, palmitoylation, phosphorylation, and disulfide bond formation.
  • The mouse Cd4 gene undergoes alternative splicing, producing isoforms that may influence function and expression.

Experimental Guidance and Technical Tips

  • This monoclonal antibody was generated using a recombinant immunogen corresponding to the extracellular region of mouse CD4 (amino acids 27–394). Flow cytometry applications targeting surface CD4 on viable cells are recommended, as the epitope is likely accessible in intact cells.
  • For flow cytometry, titrate the antibody in the relevant sample system to determine the optimal staining concentration. Include appropriate isotype controls and viability dyes to gate on live, CD4+ populations.
  • Ensure single-cell suspensions and use standard blocking procedures to minimize nonspecific binding. CD4 expression is characteristic of T-helper cells; consider co-staining with lineage markers (e.g., CD3, CD8) for comprehensive immunophenotyping.
  • The conjugate is compatible with standard 488 nm laser lines; verify compensation settings when using other fluorophores.
