  • CD4 (T-cell surface glycoprotein CD4) is also known as T-cell differentiation antigen L3T4 and T-cell surface antigen T4/Leu-3. It is an integral membrane glycoprotein essential for immune responses.
  • Functions as a coreceptor for MHC class II molecules, interacting simultaneously with the T-cell receptor (TCR) and MHC class II/peptide complexes on antigen-presenting cells.
  • Recruits the Src kinase LCK to the TCR-CD3 complex, initiating intracellular signaling pathways that lead to T-helper cell activation, lymphokine production, and adhesion.
  • In non-T cells such as macrophages and NK cells, CD4 contributes to differentiation, cytokine expression, and cell migration via a TCR/LCK-independent pathway.
  • Subcellularly localized to the cell membrane.
  • Highly expressed in T-helper cells and also present in macrophages, dendritic cells, and NK cells; CD4 expression is a hallmark of T-helper cells.
