Biological Background: CD4 Function and Localization
- CD4 (T-cell surface glycoprotein CD4) is also known as T-cell differentiation antigen L3T4 and T-cell surface antigen T4/Leu-3. It is an integral membrane glycoprotein essential for immune responses.
- Functions as a coreceptor for MHC class II molecules, interacting simultaneously with the T-cell receptor (TCR) and MHC class II/peptide complexes on antigen-presenting cells.
- Recruits the Src kinase LCK to the TCR-CD3 complex, initiating intracellular signaling pathways that lead to T-helper cell activation, lymphokine production, and adhesion.
- In non-T cells such as macrophages and NK cells, CD4 contributes to differentiation, cytokine expression, and cell migration via a TCR/LCK-independent pathway.
- Subcellularly localized to the cell membrane.
- Highly expressed in T-helper cells and also present in macrophages, dendritic cells, and NK cells; CD4 expression is a hallmark of T-helper cells.
- Post-translational modifications include disulfide bond formation, glycosylation, palmitoylation, and phosphorylation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 27-394 of mouse CD4, encompassing the extracellular immunoglobulin-like domains. This region is critical for MHC class II interaction and may be recognized in native conformation.
- Designed for flow cytometry (FC), this conjugate provides bright fluorescence suitable for multi-color panels. Titration is recommended to optimize signal-to-noise ratio.
- For surface staining, use live, unfixed cells to preserve epitope integrity. Include appropriate isotype and viability controls in experiments.
- Validate performance in your specific cell population, considering that CD4 expression levels can vary among subsets.
